Western blot - Anti-RBM15B antibody [EPR25177-158] (AB300465)
All lanes : Anti-RBM15B antibody [EPR25177-158] (ab300465) at 1/1000 dilution
Lane 1 : HeLa (Human cervix adenocarcinoma epithelial cell) transfected with scrambled siRNA control, whole cell lysate
Lane 2 : HeLa transfected with siRNA specifically targeting RBM15B, whole cell lysate 'a'
Lane 3 : HeLa transfected with siRNA specifically targeting RBM15B, whole cell lysate 'b'Lysates/proteins at 20 µg per lane.
Secondary
All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/20000 dilutionPredicted band size: 97 kDa
Observed band size: 100 kDa why is the actual band size different from the predicted?Exposure time: 114 seconds
Blocking / Diluting buffer: 5% NFDM/TBST.
Lysates were freshly made and used for Western blotting immediately to minimize protein degradation.
The expression profile/ molecular weight observed is consistent with what has been described in the literature (PMID:27602518).
The bands below 100 kDa may be caused by degradation.

Observed band size: 25 kDa why is the actual band size different from the predicted?Blocking / Diluting buffer: 5% NFDM/TBST.
This antibody does not cross-react with human RBM15.
Both recombinant proteins were expressed from E.coli expression systems.
Exposure time 7.75 seconds.
